RESUMO

INTRODUCTION: Dilated cardiomyopathy (DCM) is characterized by the enlargement of the left ventricle or biventricular, accompanied by myocardial systolic dysfunction. Chlamydia psittacosis (CP) is a zoonotic pathogen, which can cause severe pneumonia, respiratory failure, and acute organ dysfunction. The deterioration of DCM caused by CP infection is extremely rare, and few cases of successful management were reported. CASE PRESENTATION: We reported a 67-year-old male patient with DCM and chronic heart failure. Who was admitted to ICU with severe pneumonia, acute hypoxemic respiratory failure, acute decompensated heart failure, arrhythmia, and cardiogenic shock. Mechanical ventilation (MV) and venous-arterial extracorporeal membrane oxygenation (VA-ECMO) were established for respiratory and circulatory support. Broncho alveolar lavage fluid(BALF)was collected for culture and metagenomics next-generation sequencing (mNGS) test. Repeated mNGS tests indicated the high possibility of CP pneumonia, thereafter, moxifloxacin and doxycycline were prescribed. After targeted antibiotics and organ support treatment, pneumonia, respiratory and circulatory failure were gradually resolved, patient was successfully weaned from MV and VA-ECMO. Finally, the patient was recovered and discharged alive. CONCLUSIONS: Severe respiratory and circulatory failure caused by CP infection in DCM patients is a rare life-threatening clinical condition. Early accurate diagnosis, targeted antibiotic therapy, coupled with extracorporeal life support posed positive impact on the patient's disease course and outcome.

RESUMO

Land use and cover change are major factors driving global change and greatly impact terrestrial organisms, especially soil microbial diversity. Little is known, however, about bacterial diversity, distribution patterns and assembly processes across different land use types. In this study, therefore, we conducted a large-scale field survey of 48 sampling sites, encompassing different land use types in Xuancheng city, China, with different degrees of soil disturbance and different soil horizons. The distance-decay relationships (DDRs), assembly processes and the spatial patterns of soil bacterial communities were investigated based on high-throughput sequencing data. We found that the DDRs might be weakened by anthropogenic disturbances, which were not observed in tilled soils, while a decreasing trend was observed along the soil horizon in untilled soils. The relative importance of environmental factors and geographic distance varied with soil tillage. Specifically, bacterial communities in tilled soils were driven by non-spatially autocorrelated environmental factors, while untilled soils were more susceptible to geographic distance. In addition, the heterogeneity of soil properties, as well as the differences in soil bacterial niche width and niche overlap, determined the assembly processes of the bacterial community, resulting in opposite trends along the soil layers in tilled and untilled soils. These findings expand the current understanding of the biogeography of soil bacterial communities across different land use types.

RESUMO

Image super-resolution reconstruction can reconstruct low resolution blurred images in the same scene into high-resolution images. Combined with multi-scale Gaussian difference transform, attention mechanism and feedback mechanism are introduced to construct a new super-resolution reconstruction network. Three improvements are made. Firstly, its multi-scale Gaussian difference transform can strengthen the details of low resolution blurred images. Secondly, it introduces the attention mechanism and increases the network depth to better express the high-frequency features. Finally, pixel loss function and texture loss function are used together, focusing on the learning of structure and texture respectively. The experimental results show that this method is superior to the existing methods in quantitative and qualitative indexes, and promotes the recovery of high-frequency detail information.

RESUMO

A method for rapid and accurate determination of avian pathogenic Escherichia coli serotype O78 (APEC O78) by the gold nanoparticle-labeled lateral flow strip method, entitled molecule capturer analysis system (MCAS), is described. Target virulence-associated gene of APEC O78 is adopted as the analyte. After pre-amplification with the designed functional primer set, numerous new-formed amplicons are simultaneously labeled with fluorescein isothiocyanate (FITC) and digoxin. AuNPs with a diameter of 18 nm and the characteristic plasmonic peak at 526 nm are utilized for labeling. These two labels of FITC and digoxin are further captured and measured with the AuNP-labeled lateral flow strip, and the AuNPs are retained on the test line through the immunoreaction for signal output. Under optimized conditions, this MCAS protocol can determine the target APEC O78 with excellent determination limit of 4.3 cfu mL-1 based on the optical density of AuNPs on the test line of lateral flow strips. The working range is 2.52 × 101 to 1.63 × 107 cfu mL-1. Spiked serum samples are rapid and accurately measured, and the results are highly correlated with those of the real-time PCR. With this MCAS protocol, rapid and on-site determination of APEC O78 can be realized without expensive instruments or professional personnel. This MCAS protocol can be easily applied to other analytes by just replacing the traditional primer set with functionalization primer set.
